November 2011 wird Prof. Dr. Amos Tanay (Weizmann Institute, Department of Computer Science and Applied Mathematics) im Rahmen der ZBI "Distinguished Speaker Series" einen Vortrag zum Thema "The evolution of normal and aberrant epigenomes" halten (Raum 001, ZBI (Gebäude E 2.1); Beginn: Punkt 17:00 Uhr).



What comes first – regulatory or epigenetic change?  Epigenetic changes are frequently observed during normal development and in cancer cells. The regulatory role of these changes is difficult to characterize, since the epigenomic state is strongly coupled to the genome's activity patterns and since most experiments cannot follow the dynamics of this coupling in high temporal resolution.  I will present several projects that are running in the lab, which aim to use experimental and theoretical evolution in order to follow together changes in DNA methylation, histone modifications, chromosomal architecture and genomic sequences. One outcome of these experiments is a working model for understanding how specific cancer DNA methylation patterns are emerging. The model involves a continuous flux of stochastic small changes that are restricted by competing epigenetic factors including Polycomb repressive complexes, CTCF and the core transcriptional machinery. Finally, I'll present data from new high resolution experiments (4C-seq, Hi-C) quantifying contact intensities between remote chromosomal loci. Models for interpreting dynamic epigenomic states given these exciting new snapshots of nuclear architecture will be discussed.…
